Hexapeptide-48 HCl is a synthetic peptide that acts as a specific agonist for the delta opioid receptors. Expression of the delta opioid receptor is modulated in inflammatory conditions and under stress. It has the sequence Tyr–Pro–Leu–Asp–Leu–Phe and is known under the trade name Rubixyl®.

Inflammatory stress leads to the dysregulation of the expression of genes and proteins involved in the maintenance of epidermal integrity and the barrier function of the epidermis. Hexapeptide-48 HCl is an anti-stress peptide and is capable of blocking the depletion of the delta opioid receptor in inflammatory conditions.

Topical application of the preparation containing Hexapeptide-48 HCl regulates the expression of regulated genes and proteins. In addition, it diminishes the influence of chronological stress aging and cigarette smoke-induced skin stress.

In vitro and in vivo studies showed that activation by a natural agonist of the delta opioid receptor Hexapeptide-48 HCl reduces skin inflammation while improving epidermal differentiation and barrier function. The study of healthy volunteers confirmed the effectiveness of the Rubixyl peptide in repairing damaged skin by reducing TEWL, increasing hydration, and reducing wrinkles, including in the perioral (around the mouth) and ocular (around the eyes) areas.

Hexapeptide-48 HCl is used in high-end anti-aging, anti-wrinkle, and eye-care products in concentrations of 0.1-2%.
