Myristoyl Tetrapeptide-6 is a synthetic lipopeptide that inhibits human neutrophil elastase (HSE) and exhibits anti-aging properties. It has the sequence Myr-Ala-Ala-Pro-Val (mAAPV) and is known under the trade name DermaPep™ A420.

Transforming growth factor (TGF-β), a member of a large family of secreted peptide growth factors, plays a significant role in ECM (extracellular matrix) production by increasing the expression of ECM genes, including collagen, elastin, fibronectin, and proteoglycans. According to the manufacturer's claim, Myristoyl tetrapeptide-6 mimics TGF-β, acts via the TGF-β signaling pathway, and thereby up-regulates ECM proteins such as collagen and fibronectin.

The motif Ala-Ala-Pro-Val and its derivatives are known inhibitors of serine proteases, particularly human neutrophil elastase, an enzyme that can degrade dermal elastin. Independent studies showed that lipidation of Ala-Ala-Pro-Val increases its stability and enables transdermal delivery. Myristoyl Tetrapeptide-6 inhibits HSE, preventing elastin degradation and helping preserve elastic fibers.

The manufacturer's in vitro and in vivo results show that Myristoyl Tetrapeptide-6 significantly increases the expression of type I procollagen and fibronectin, efficiently reduces fine lines and wrinkles, and helps maintain dermal thickness. It can be incorporated in cosmetic formulations such as emulsions, oily serums, gels, and creams for anti-aging and anti-wrinkle purposes.
