Phytosan™ is a botanical complex, an active prepared from soy seeds, containing proteins, glycoproteins, and polysaccharides. It protects against light-induced skin aging and UV-induced energy loss, slows down the skin‘s aging process, and boosts DNA condition in epidermal stem cells, thus ensuring lasting high epidermal quality. Aging makes the skin thinner, drier, and more delicate.

Phytosan™ is a cosmetic active ingredient that stimulates the skin’s natural protection function after UV irradiation and can be defined as a biological UV protector. It contributes to the sun protection factor (SPF) without UV absorption capacity. In the meantime, using SPF as an indicator of sun protection is being considered. The SPF reflects only the UV-induced erythema.

Other consequences of UV exposure are photoallergies, photoaging, immunosuppression, and cancer. The introduction of immunoprotection factors (IPF) or mutation-preventing factors (MPF) apart from SPF has already been proposed. Undoubtedly, Phytosan™ is acting as a mutation-preventing factor due to the proven activation and stabilization of p53.

This complex was investigated for its influence on the endogenous protective mechanism after chronic and single UV irradiation of human keratinocytes. It was shown that Phytosan™ in different concentrations could stimulate and stabilize p53 tumor suppressor protein by p14ARF activation. This explains the additionally measured increase in DNA repair effectiveness and the reduction of non-repairable cell formation (apoptotic or sunburn cells). Specifically designed tests on humans have proven that the induction of natural cellular protective functions leads to a higher UV tolerance in vivo and to the counteraction of premature skin aging.

Mechanism of action

Sunburn, an inflammatory response, is the most apparent sign of excessive UV exposure to the skin. Other critical impacts of the sun are erythema, photoallergies, photoaging, immunosuppression, and cancer.

Phytosan™, a soy seed-derived active, was a potent activator of the tumor suppressor protein p53 detected after single and chronic UV irradiation of primary human keratinocytes. Furthermore, it stimulates the expression of the tumor suppressor p14ARF, which inactivates the oncogene Mdm2, a p53 antagonist.

After phosphorylation, p53 acts as a transcription factor, binding specifically to other genes and controlling expression. The p21 protein organizes cell cycle arrest in the case of DNA damage; the gadd45 protein activates DNA repair, and the Bax protein induces apoptosis of non-repairable cells. This functionality makes p53 the most crucial tumor suppressor gene family member.

Depending on the activation and stabilization of p53, it could also be shown that treating keratinocytes with Phytosan™ leads to significantly increased DNA repair effectiveness and prevents the formation of non-repairable apoptotic cells. The significance of the induction of cellular protective functions by Phytosan™ in vitro could also be confirmed in studies on humans.

Treatment of the skin with a Phytosan™-containing formulation increases its UV tolerance, which can be expressed in SPF units. In addition, the active prevents the UV-induced release of proteases and thus prevents the destruction of extracellular matrix compounds. It stimulates the skin's natural defense system and can be defined as a biological UV protector that prevents severe photodamage and accelerated skin aging.
